Overview of Research

Non-invasive fluorescence molecular imaging will allow clinicians to visualize the expression and activity of specific biomolecule which is not possible with the existing imaging techniques such as X-ray computed tomography (CT) and nuclear magnetic resonance imaging (NMRI). Fluorescence imaging in the shortwave-infrared (SWIR) region (1000 nm – 1400 nm) can be used to perform deep biological tissue penetration. The absorption and scattering of SWIR light in the biological tissue is very small and almost negligible autofluorescence. The fluorescence imaging at this region provides much higher signal to background ratio compared with the conventional near-infrared (NIR) region 700 nm – 900 nm). Upon successful development of SWIR-emitting molecular probes recently, we demonstrated targeted breast cancer imaging. Currently, we are developing cyanine fluorophores with aqueous solubility, higher stability, and improved quantum yield/fluorescence brightness for the possible clinical optical diagnostics. On the other hand, exosomes are naturally occurring membranous vesicles with unique and important pharmacological properties, secreted by various types of cells. Even though the exosomes were reported to facilitate the uptake of Aβ by microglia, its mechanism is not completely know. We are trying to understand the exosome biology in normal and disease condition through in-vivo tracking of exosomes by SWIR fluorescence imaging. Also, we are developing small molecule modulators to enhance exosome production to discover potent drug candidate for Aβ clearance and Alzheimer’s disease.
